dc.description.abstractEl objetivo general de esta tesis fue contribuir al conocimiento de la fisiología del sistema reproductor masculino a través del estudio de cómo esfingolípidos que son específicos de las células espermatogénicas (o células germinales, CG) cambian con el avance de la maduración sexual y la diferenciación celular. El foco estuvo sobre las especies moleculares de esfingomielina (SM) y de ceramida (Cer) con ácidos grasos poliinsaturados (PUFA) de muy larga cadena (VLCPUFA), que se presentan como VLCPUFA no hidroxilados (n-V) y 2-hidroxilados (h-V) de hasta 32 átomos de carbono en las CG de rata. Los objetivos específicos fueron i) investigar cómo se distribuyen estas especies moleculares entre las membranas de las CG, ii) determinar cómo cambia con el desarrollo testicular y la diferenciación celular la expresión de los genes que codifican a las enzimas responsables de la biosíntesis de los n-V y los h-V, iii) establecer si las CG aisladas son capaces de biosintetizar por sí mismas estas especies de Cer y de SM entre otras a través de la vía de novo y de expresar las correspondientes sintasas, y iv) inquirir sobre factores endocrinos y/o paracrinos capaces de modular dicha actividad y/o expresión. Los efectos del desarrollo se estudiaron en testículos de animales de distintas edades postnatales (P), y los de la diferenciación en poblaciones de CG en los estadios de espermatocitos en paquiteno (EP), espermátidas redondas (ER), y espermátidas elongadas o tardías (ET), aisladas a partir de las células totales (CT) del epitelio seminífero de ratas adultas. En algunas mediciones se incluyeron a los cuerpos residuales (CR) y a las células de Sertoli (SC). Las poblaciones de CG se aislaron utilizando gradientes continuos de albúmina. Los estudios que requirieron separación, aislamiento e identificación de clases de lípidos y especies moleculares se hicieron por técnicas cromatográficas de alta resolución y sensibilidad. Las investigaciones sobre biosíntesis de n-V o de esfingolípidos (SL) se hicieron en CG mantenidas en cultivo primario, siguiendo las transformaciones biológicas de sustratos radioactivos. La expresión de genes a nivel ARNm se comparó aplicando PCR cuantitativa, y a nivel proteína se evaluó utilizando técnicas de Western blot seguida de inmunomarcación con anticuerpos, y en cortes de tejido por microscopía de inmunofluorescencia. En la primera parte de la tesis se compararon la localización y la distribución de glicerofosfolípidos (GPL), colesterol y SM entre fracciones de membrana obtenidas de EP, ER y ET. El foco estuvo puesto en la distribución lateral de las especies moleculares de SM y Cer con VLCPUFA entre fracciones de membrana conteniendo dominios tipo raft y no-raft, partiendo de la hipótesis de que estas especies se verían excluidas de los primeros. Utilizando preparaciones de CT, inicialmente comparamos dos métodos, originalmente diseñados para estudiar la separación lateral de proteínas en membranas, para decidir cuál sería más apropiado para evaluar la distribución lateral de los lípidos. El método clásico, que utiliza detergente en frío para obtener fracciones de membrana ricas en dominios tipo raft, mostró bajas recuperaciones de lípido y proteína, separación incompleta de los lípidos entre dominios, interferencia del detergente en los análisis, e hidrólisis parcial de GPL y SM. Se encontró preferible un método que usa gradientes de sacarosa para separar fracciones de membrana sobre la base de su densidad a partir de homogenados totales (HT), pues permitió separar fracciones de membrana livianas, pesadas, y extra-pesadas (ML, MP y MEP, respectivamente) además de una fracción aún más pesada en el pellet o sedimento de la homogenización. La recuperación, tanto del fósforo lipídico como de la proteína, alcanzó, en promedio, un 85% del originalmente presente en los HT. Proteínas marcadoras mostraron que las pequeñas ML contenían dominios tipo-raft/caveolas, y las abundantes MP dominios no-raft, principalmente derivados de la membrana plasmática, mientras las MEP eran ricas en membranas intracelulares. Consistente con su rol como precursoras de esfingolípidos con VLCPUFA, especies de Cer con n-V y h-V se hallaron más concentradas en las MEP. Como se esperaba, las ML contenían GPL y SM con AG saturados, mientras las MP eran ricas en GPL con PUFA y SM con VLCPUFA. Llamativamente, tal vez debido a la alta insaturación de esos ácidos grasos, la relación colesterol/fosfolípidos fue más alta en las MP que en las ML. Con el avance de la diferenciación, el contenido de Cer con VLCPUFA, especialmente h-V Cer, tendió a aumentar en las MP de las CG en el sentido EP→ER→ET. En el mismo sentido, mientras los ácidos grasos de los lípidos de ML no variaron, en los de las MP las relaciones 22:5n-6/20:4n-6 en GPL y h-V/n-V en SM y en Cer aumentaron en forma altamente significativa. En la segunda parte, se determinó la expresión de enzimas que se espera jueguen un papel en la biosíntesis de los n-V y los h-V de SM y Cer. El foco estuvo puesto en la Elovl4 y la Fa2h, con la hipótesis, basada en lo que habían mostrado los lípidos, de que sus expresiones aumentarían con el desarrollo postnatal en el testículo y que, en las CG de animales adultos, variarían en sentidos opuestos, la primera disminuyendo y la segunda aumentando con la diferenciación. Seis de los 7 miembros de la familia de genes Elovl, se expresaron a nivel ARNm en CG. Los genes Elovl5 y Elovl2 se incluyeron en el estudio por codificar elongasas cuya actividad parcialmente se superpone, colaborando en la biosíntesis de PUFA, y porque Elovl2 es esencial en la formación de PUFA de 24 carbonos que sirven de sustrato a la Elovl4. Contrario a lo esperado, el nivel de ARNm de Elovl4 se halló elevado en testículos que virtualmente carecían de CG (y de SM o Cer con n-V), como lo fueron los de animales en edades infantiles (P14) y los de adultos (P90) que habían sido despojados de CG por exposiciones repetidas a episodios de hipertermia. Esta aparente inconsistencia se debió a que el nivel de ARNm de Elovl4 fue inesperadamente alto en las SC. Con la maduración sexual, los niveles de Elovl4-ARNm en el testículo decrecieron hasta la adultez, para permanecer luego relativamente constantes. Entre las células espermatogénicas del adulto, los contenidos más altos de este ARNm aparecieron en las ET y en los CR. En contraste con Elovl4, Fa2h no se expresó como ARNm en el testículo hasta la edad de aparición de las ER (P26), y sus niveles aumentaron con el crecimiento y la diferenciación celular. Como proteínas, ni Elovl4 ni Fa2h se expresaron en las SC. Elovl4 estuvo más concentrada en EP que en ER, en las que mostró una localización perinuclear. En el citoplasma de las ET apareció concentrada en pequeñas estructuras esféricas que podrían ser CR en formación. Como proteína, Fa2h se encontró especialmente concentrada en las ET, asociada a una estructura supranuclear que juega un rol importante en dar la forma definitiva a la cabeza de los futuros espermatozoides. Elovl4 y Fa2h no se detectaron en las gametas liberadas desde el epitelio seminífero. La actividad combinada de las elongasas en estudio (Elovl5, Elovl2 y Elovl4) se evaluó comparando las conversiones sufridas por el [3H]20:4n-6 en cultivos de EP, ER y ET, así como de SC. Este PUFA fue elongado en las cuatro poblaciones celulares a [3H]22:4 y [3H]24:4, y estos productos fueron activamente desaturados a [3H]22:5 y [3H]24:5. Como se esperaba, se encontraron n-V tetraenoicos y pentaenoicos marcados con [3H] de 26 y hasta 32 carbonos sólo en las CG. La actividad de elongación de PUFA decreció en el sentido EP→ER→ET. En la tercera parte, con la hipótesis de que las CG serían capaces de producir sus propios esfingolípidos, investigamos cómo espermatocitos y espermátidas biosintetizan Cer, SM y GlcCer, y cómo dicha actividad, así como la expresión de los genes de las correspondientes sintasas, cambian con la diferenciación. Paralelamente evaluamos la posibilidad de que la biosíntesis y/o la expresión génica estuvieran afectadas por la testosterona (Tes) y por factores solubles liberados desde las SC. Empleando como precursor al [3H]16:0 y dos inhibidores específicos de la ruta biosintética de novo (Lcicloserina y fumonisina B1) hallamos que las CG, aisladas y en cultivo, son capaces de biosintetizar sus propias Cer y SM, incluyendo sus especies moleculares con VLCPUFA, además de GlcCer. Estas actividades biosintéticas fueron máximas en los EP, disminuyendo con la diferenciación (EP>>ER). La Tes estimuló la biosíntesis de [3H]SM sólo en las ER. La suplementación con el medio condicionado obtenido de cultivos de SC (MCS) estimuló significativamente la formación de [3H]Cer tanto en EP como en ER, la de [3H]SM sólo en ER, y la de [3H]GlcCer sólo en EP. La Tes y el MCS incrementaron a todas las especies moleculares de Cer y de SM, incluyendo las n-V. En comparación con el MCS sólo, la combinación Tes+MCS promovió significativamente la biosíntesis de [3H]SM en los EP, mientras la redujo en las ER, en las cuales incrementó la marcación de [3H]Cer y de [3H]GlcCer. El estímulo sobre la marcación de Cer y SM fue específico para las [3H]n-V Cer y [3H]n-V SM. El significativo fomento que sobre la biosíntesis de novo ejerció el MCS indica que factores solubles (o transportados en microvesículas) provenientes de las SC (incluyendo tal vez distintas formas de ARN), juegan un rol en promover, en las CG, la biosíntesis de esfingolípidos destinados a sus membranas. En esta tercera parte de la tesis también se investigó la expresión a nivel ARNm de genes que codifican para cuatro sintasas (S) clave implicadas en la biosíntesis de SL (CerS3, SMS1, SMS2, y GlcCerS), de tres elongasas de ácidos grasos (Elovl5, Elovl2 y Elovl4), y de Fa2h. Se encontró que los niveles de ARNm de CerS3 fueron máximos en EP y los de SMS2 máximos en ER, ambos disminuyendo en las ET. Los de GlcCerS disminuyeron menos que éstos con la diferenciación, y los de SMS1 no variaron. Los cuatro ARNm también aparecieron en los CR. Mientras CerS3 no se expresó en las SC, los ARNm de las demás sintasas se encontraron en ellas, aunque en cantidades menores que en las CG. En la última parte de esta sección se estudiaron, en las células espermatogénicas totales, los efectos de la Tes, del MCS, y de Tes+MCS sobre los niveles de ARNm de las enzimas mencionadas. Las expresiones como ARNm de Elovl2 y Elovl4 mostraron ser reguladas por el MCS, las de SMS2 y Elovl5 por MCS y Tes, y la de Fa2h sólo por Tes. La Tes y el MCS, tanto por separado como combinados, no afectaron los niveles de ARNm de CerS3, SMS1, o GlcCerS, mientras tendieron a reducir los de SMS2. La presencia de Tes no afectó el ARNm de Elovl2 ni el de Elovl4, mientras tendió a aumentar el de Elovl5. El MCS, por su parte, redujo significativamente los ARNm de las tres elongasas. Cuando Tes y MCS se combinaron, el ARNm de Elovl5 aumentó, y los de Elovl2 y Elovl4 continuaron bajos, recapitulando la influencia de Tes y del MCS, respectivamente. El efecto más significativo de la Tes, tanto sola como combinada con el MCS, fue el de estimular la expresión de Fa2h. Tomados en conjunto, estos resultados muestran por primera vez que factores que juegan roles fisiológicos en la espermatogénesis, como lo son la testosterona y moléculas liberadas desde las células de Sertoli, son capaces de estimular la biosíntesis de novo de esfingolípidos en las células espermatogénicas y de regular, en algunos casos a la alta y en otros a la baja, la expresión génica de algunas de las enzimas responsables de dicha biosíntesis.es
dc.description.abstractThe overall objective of this work was to contribute to the knowledge of the physiology of the male reproductive system by examining how molecular species of sphingolipids that are specific of spermatogenic cells (or germ cells, GC) change with the progress of sexual maturation and cell differentiation. The focus was on the sphingomyelins (SM) and ceramides (Cer) having polyunsaturated fatty acids (PUFA) with very long chains (VLCPUFA), which occur in their non-hydroxy (n-V) and 2-hydroxy (h-V) forms and have up to 32 carbon atoms in rat GC. The specific aims were i) to investigate how these molecular species distribute among the GC membranes, ii) to determine how the expression of genes that code for the enzymes responsible for the biosynthesis of n-V and h-V changes with testis maturation and GC differentiation; iii) to ascertain whether isolated GC are able to biosynthesize these species of Cer and SM among others through the de novo pathway and to express the corresponding synthases, and iv) to inquire into endocrine and/or paracrine factors that are capable of modulating such an activity and/or expression. The effects of development were studied in testes at specific postnatal ages, and those of differentiation in GC populations at the stages of pachytene spermatocytes (PtS), round spermatids (RS), and elongated or late spermatids (LS) that were isolated from the total cells (TC) of the seminiferous epithelium of adult rats. In some analyses the residual bodies (RB) and the Sertoli cells (SC) were included. The GC populations were isolated using continuous gradients of albumin. The studies that required separation, isolation, and identification of lipid classes and molecular species were performed using chromatographic techniques of high resolution and sensitivity. The investigations about biosynthesis of n-V and of sphingolipids (SL) were done in GC maintained in primary culture by following the biological transformations of radioactive substrates. Gene expression at the mRNA level was compared by applying quantitative PCR, and at the protein level it was evaluated using Western blot followed by immunolabeling with antibodies, and in tissue sections by immunofluorescence microscopy. In the first part of the thesis the localization and distribution of glycerophospholipids (GPL), cholesterol, and SM among membrane fractions obtained from PtS, RS, and LS were compared. The focus was on the lateral distribution of the molecular species of SM and Cer with VLCPUFA between membrane fractions containing raft-like and non-raft domains, with the hypothesis that these species would be excluded from the former. Using TC preparations, initially we compared two methods, originally designed to study the lateral separation of proteins in membranes, in order to decide which one would be most suitable to evaluate the lateral distribution of lipids. The classic method that uses detergent at low temperature to obtain fractions rich in raft-like domains showed poor recuperation of lipid and protein, incomplete separation of lipids between membrane fractions, interference of the detergent in lipid analyses, and partial hydrolysis of GPL and SM. A method that employs sucrose gradients to separate membrane fractions from total homogenates (TH) on the basis of their densities was preferred, as it allows separation of light, heavy, and extra-heavy membrane fractions (LM, HM, and EHM, respectively) and an even heavier fraction in the pellet or sediment of homogenization. The recovery of lipid phosphorus and protein amounted, in average, to an 85% of that originally present in the TH. Protein markers showed that the small LM fractions contained raft-like/caveolae domains, and that the abundant HM had non-raft domains, mostly derived from cell plasma membranes, whereas the EHM were rich in intracellular membranes. Consistent with their role as precursors of sphingolipids with VLCPUFA, Cer species with n-V and h-V were found most concentrated in EHM. As expected, the LH contained GPL and SM with saturated fatty acids, whereas the HM fractions were rich in GPL with PUFA and SM with VLCPUFA. Interestingly, perhaps because of the high unsaturation of these fatty acids, the cholesterol/phospholipid ratio was higher in HM than in LH. The content of Cer with VLCPUFA, especially h-V Cer, tended to increase in the MP of GC with the progress of differentiation in the direction PtS→RS→LS. In the same direction, the fatty acids of LH lipids did not vary, while in those of MP the 22:5n-6/20:4n-6 ratio in GPL and the h-V/n-V ratio in SM and Cer increased in a highly significant form. In the second part, the expression of enzymes that are expected to play a role in the biosynthesis of the n-V and h-V of SM and Cer was determined. The focus was placed on Elovl4 and Fa2h, on the hypothesis, based on what lipids had shown, that their expressions would increase in testes with postnatal development and that, in adult animal GC, their expressions would vary in opposite directions, the former decreasing and the latter increasing with cell differentiation. Six of the 7 members of the Elovl gene family were expressed at the mRNA level in GC. The Elovl5 and Elovl2 genes were included in the survey as they code for elongases with partially overlapping activities, collaborating in the biosynthesis of PUFA, and because Elovl2 is essential in the formation of the 24 carbons PUFA that serve as substrates to Elovl4. Against our expectations, the Elovl4 mRNA level was found high in testes virtually lacking GC (and SM or Cer with n-V), as were those of animals in infantile ages (P14) and those of adults that had been deprived of their GC by repeated exposures to episodes of hyperthermia. This apparent inconsistency was due to the fact that the mRNA level of Elovl4 was unexpectedly high in SC. With sexual maturation, the Elovl4-mRNA levels in testes decreased up to adulthood, then remaining relatively constant. Among adult spermatogenic cells, the highest content of this mRNA appeared in LS and RB. In contrast with Elovl4, Fa2h did not express itself as mRNA in testes until the appearance of RS (P26), and their levels increased with cell growth and differentiation. As proteins, Elovl4 and Fa2h were not expressed in SC. Elovl4 showed a perinuclear localization, was more concentrated in PtS than in RS, and then appeared concentrated in small spherical structures in LS cytoplasm, which could be RB in formation. As a protein, Fa2h was found mainly concentrated in LS, associated to a supranuclear structure that plays an important role in giving the final shape to the heads of the sperm cells to-be. Elovl4 and Fa2h were not detected in the gametes released from the seminiferous epithelium. The combined activity of the elongases under study (Elovl5, Elovl2 and Elovl4) was evaluated by comparing the conversions undergone by [3H]20:4n-6 in cultures of PtS, RS and LS, as well as of SC. In the four cell populations this PUFA was elongated to [3H]22:4 and [3H]24:4, and these products were actively desaturated to [3H]22:5 and [3H]24:5. As expected, [3H]-labeled tetraenoic and pentaenoic n-V having 26 to 32 carbons were found only in GC. The activity of PUFA elongation decreased in the direction PtS→RS→LS. In the third part, with the hypothesis that GC would be able to produce their own SL, we investigated how spermatocytes and spermatids biosynthesize Cer, SM and GlcCer, and how such an activity, as well as the expression of genes of the corresponding synthases, would change with cell differentiation. In parallel we evaluated the possibility that biosynthesis and/or gene expression would be affected by testosterone (Tes) and by soluble factors released from SC. Employing [3H]16:0 as precursor and two specific inhibitors of the de novo biosynthetic pathway (L-cycloserine and fumonisin B1), we found that GC, isolated and in culture, are able to biosynthesize their own Cer and SM, including their molecular species with VLCPUFA, and also GlcCer. These biosynthetic activities were maximal in PtS, decreasing with differentiation (PtS>>RS). Tes stimulated the biosynthesis of [3H]SM, only in RS. Supplementation with the conditioned medium obtained from SC cultures (CMS), significantly stimulated the formation of [3H]Cer in PtS and RS, that of [3H]SM only in RS, and that of [3H]GlcCer only in PtS. All the molecular species of Cer and SM were increased by Tes and CMS, including the n-V. In comparison with CMS, the combination Tes+CMS significantly promoted the biosynthesis of [3H]SM in PtS, while reducing it in ER, in which cells it increased the labeling of [3H]Cer y de [3H]GlcCer. The stimuli on Cer and SM labeling was specific on the [3H]n-V Cer and [3H]n-V SM. The significant enhancement that CMS exerted on the de novo biosynthesis indicates that factors that are soluble (or transported in microvesicles) coming from SC (perhaps including different forms of RNA), play a role in promoting in CG the biosynthesis of SL destined to their membranes. In this third part of the thesis we also evaluated the expression, at the mRNA level, of genes that code for four key sinthases (S) involved in the biosynthesis of SL (CerS3, SMS1, SMS2, and GlcCerS), of three fatty acid elongases (Elovl5, Elovl2 y Elovl4), and of Fa2h. We found that the mRNA levels of CerS3 were maximal in PtS and those of SMS2 maximal in RS, while both decreased in LS. Those of GlcCerS varied less than these with differentiation and those of SMS1 did not vary. The four mRNA also appeared in RB. Whereas CerS3 was not expressed in SC, the mRNA of the other synthases was, although in lesser amounts than in GC. In the last part of this section the effects of Tes, CMS, and Tes+CMS on the mRNA levels of the above-mentioned enzymes were studied in total spermatogenic cells. The expressions as mRNA of Elovl2 and Elovl4 were regulated by CMS, those of SMS2 and Elovl5 by CMS and Tes, and that of Fa2h only by Tes. Tes and CMS, either separately or in combination, did not affect the mRNA levels of CerS3, SMS1, or GlcCerS, while tended to reduce those of SMS2. The presence of Tes did not affect the mRNA of Elovl2 or Elovl4, while tended to increase those of Elovl5. The CMS reduced significantly the mRNA levels of the three elongases. When Tes and CMS were together, the mRNA of Elovl5 increased, and those of Elovl2 and Elovl4 continued low, recapitulating the influence of Tes and CMS, respectively. The most significant effect of Tes, either alone or combined with CMS, was to stimulate the expression of Fa2h. Taken together, these results show for the first time that factors that play physiological roles in spermatogenesis, as testosterone and molecules released from Sertoli cells do, are able to stimulate the de novo biosynthesis of sphingolipids in spermatogenic cells, and to regulate, in some cases positively and in others negatively, the gene expression of some of the enzymes responsible for such biosynthesis.es
